In English they are kind of same thing.
Sinä et voi tehdä sitä minulle = You can't to that to me
Hän ei ole voinut tietää siitä/Hän ei voinut tietää siitä = He/She couldn't have known that.
Both are THAT but it really depends on the sentence

The difference is quite difficult to explain though I'm sure there is some grammatical explanation for it 🤔
I think "siitä" can be translated translated as "about that"/"about that thing" while "sitä" is "that thing", if that makes sense.

Sitä = se in partitive form
Siitä= se in elative form
In the first sentence you must use the partitive form of "se", because the sentence is in negative. The partitive is used here to express direct object and accusative can't be used because the sentence is in negative.
In the second sentence both sitä and siitä could be used. The difference isn't great. "Hän ei ole voinut tietää sitä" sounds like he/she couldn't have known a singular thing or a fact whereas the version with "siitä" sounds like he/she couldn't have known something about a certain subject. I try to give examples:
1. "Tv-visassa kysyttiin kuka oli Yhdysvaltojen kolmas presidentti. En tiennyt sitä!" = They asked in a game show who was the third president of the United States. I didn't know it!
2. "Isäni oli ostanut uuden moottoripyörän. En tiennyt siitä!" = My father had bought a new motorcycle. I didn't know (about) that!
So compare them:
The third president of the USA = the president is a single person
My dad had bought a new motorcycle = a whole sentence, a wider subject
